Correct Connection Method for LC Spectrum Splitter

To correctly connect an LC spectrum splitter, use appropriately cut PEEK tubing to achieve the desired split ratio, connect the shorter tubing to waste, the longer tubing to the detector or MS, and configure the software to manage flow.

Step-by-Step Connection

  1. Determine Split Ratio Decide the desired flow split between the detector and waste. For example, a 1:3 split means 25% of the flow goes to the detector and 75% to waste.
  2. Cut PEEK Tubing Measure and cut the PEEK tubing according to the split ratio. For a 60 cm total length, cut 20 cm for the detector side (1/3) and 40 cm for the waste side (2/3) to maintain the correct flow distribution .
  3. Connect to Diverter Valve
    • Disconnect the tubing from port 3 of the fluidics diverter valve.
    • Connect the shorter tubing to port 3 and place the outlet in a waste container.
    • Connect the longer tubing from port 2 to the detector, such as an ESI probe or optical detector .
  4. Software Configuration Configure the acquisition software to recognize the split flow. Ensure that flow to waste and flow to the detector are properly managed, and that the disconnected channel is not used for calibration or post-column addition .
  5. Prevent Overpressure When connecting directly to UV, PDA, or FLR detectors, splitting the flow helps avoid overpressure in the flow cell. Always verify that the tubing and connectors are compatible with the mobile phase and system pressure .
  6. Maintenance and Verification After installation, flush the system with blank runs to remove any air bubbles or particulates. Perform a test injection to confirm that the split ratio is correct and that all peaks are properly detected .

Additional Tips

  • Use high-quality, compatible PEEK tubing to prevent leaks or contamination.
  • Ensure all unused ports are properly terminated to avoid backpressure or flow disturbances.
  • Avoid cascading too many splitters in series, as this can reduce signal quality and increase system pressure . Following these steps ensures accurate flow splitting, protects detectors, and maintains chromatographic performance.
