Uvvisible Spectrometer

A UV-Vis spectrometer measures the absorption of ultraviolet and visible light by a sample to analyze its chemical composition and concentration.

Principle of Operation

A UV-Vis spectrometer works on the principle of absorption spectroscopy, where a photon of light is absorbed by a molecule, promoting an electron from a lower energy state to a higher energy state . The amount of light absorbed at each wavelength is proportional to the concentration of the absorbing species, following the Beer-Lambert law. By scanning across the UV (190–400 nm) and visible (400–800 nm) regions, the instrument generates an absorbance spectrum unique to each compound .

Components

Key components of a UV-Vis spectrometer include :

  • Light Source: Typically a deuterium lamp for UV and a tungsten lamp for visible light.
  • Monochromator: Selects specific wavelengths of light to pass through the sample.
  • Sample Holder/Cuvette: Holds liquid, solid, or gas samples.
  • Detector: Measures transmitted or reflected light to calculate absorbance or transmittance.
  • Data Processor: Converts detector signals into spectra for analysis.

Measurement Parameters

  • Absorbance (A): Indicates how much light is absorbed by the sample.
  • Transmittance (%T): The fraction of light passing through the sample.
  • Wavelength (nm): Determines which electronic transitions are being measured .

UV-Vis spectrometers are widely used in:

  • Chemistry and Biochemistry: Quantifying nucleic acids, proteins, and transition metal ions.
  • Environmental Science: Monitoring pollutants and water quality.
  • Pharmaceuticals: Assessing drug purity and concentration.

Limitations

  • Less sensitive compared to fluorescence or mass spectrometry.
  • Requires the sample to have a chromophore that absorbs in the UV-Vis range.
  • High concentrations may lead to deviations from Beer-Lambert law . UV-Vis spectrometers remain a fundamental analytical tool in laboratories due to their simplicity, broad applicability, and ability to provide both qualitative and quantitative information about molecular species.
